地塞米松磷酸钠壳聚糖-海藻酸钠微囊的制备工艺研究 被引量:7

摘要 目的探讨壳聚糖海藻酸钠微囊用于运载地塞米松磷酸钠的新方法。方法采用凝聚法制备了地塞米松磷酸钠壳聚糖海藻酸钠微囊,以包封率、微囊强度为指标,设立总的评估指数,选取海藻酸钠浓度、壳聚糖浓度、CaCl2浓度和地塞米松磷酸钠用量四个因素,每个因素选取三个水平,选用L9(34)表进行正交实验优化制备工艺。结果四个因素中,壳聚糖浓度对评估指数的影响最大(P<0.01),其次是CaCl2和海藻酸钠浓度(P<0.05),地塞米松磷酸钠对评估指数的影响没有统计学意义(P>0.05)。按照优化方案进行正交实验复核实验表明,药物包封率、微囊机械强度分别为96.90%,97.33%。结论本法工艺简便、稳定,具有应用前景。 OBJECTIVE To study the new preparation of dexamethsone sodium phosphate(DSP)chitosan-alginate microencapsules. METHODS Dexamethsone sodium phosphate chitosan-alginate microencapsules(DSP-CAM)were prepared by coacervation technology.Four factors,including alginate,chitosan,CaCl_ 2 and DSP, and three levels for each factor that may affect the formulation of microencapsules were selected and arranged in an L_9(3 4 ) orthogonal experimental table.The evaluating indicator(EI%),calculated according to drug encapsulated efficiency(EE%) and strength of microencapsules(MS%),was introduced as a total index for the microencapsules formulation.RESULTS The four factors affected EI differently.Chitosan behaved as the maximum( P <0.01),followed alginate and CaCl_ 2 ( P <0.05).However,the DSP did not influence EI( P >0.05).The orthogonal experiment was checked up and the result indicated that EE% and MS% were 96.90% and 97.33% respectively.CONCLUSION The preparation procedure is good,simple and reproducible.
