中药牛膝提取物抗肿瘤活性的初步研究

The research of extract of Achyranthes Bidentata Blume′s anti-tumor activity

目的 探讨中药牛膝提取物的抗肿瘤活性及其作用机制。方法 MTT法检测细胞抑制率;流式细胞仪分析细胞周期及凋亡情况;分别用生物化学法和ELISA法检测巨噬细胞因子TNF α和IL- 6的表达;用RT -PCR法检测巨噬细胞因子IL 6mRNA表达。结果 牛膝提取物在体外对两种肿瘤细胞株的增殖具有明显抑制作用,量效、时效关系显著(P <0 .0 1) ,细胞周期停滞于G0 G1 期,诱导细胞凋亡可能是其发生作用的机制之一;牛膝提取物虽然在体外不能促进小鼠脾细胞的生长增殖,但对巨噬细胞的吞噬功能却有较强的刺激作用(P <0 .0 1) ,并可促进细胞因子TNF -α和IL- 6的产生以及IL- 6mRNA的表达。结论 该提取物具有抗肿瘤作用,不仅对免疫功能没有抑制作用,而且可以通过延缓肿瘤细胞周期、诱导凋亡、增强巨噬细胞对肿瘤细胞的杀伤作用及分泌细胞因子如TNF- α、IL -6等参与其抗肿瘤机制。

Objective To investigate the antitumor effect an d mechanism of extract of Achyranthes Bidentata Blume(eAbB). Methods The effects of different doses′s eAbB on BGC823 cell, K562 cell ac tivity were detected by MTT method; the cell cycle was measured by FACS; the pro duction of TNF-α and IL-6 were measured by bio-chemical method and ELISA res pectively; the expressions of IL-6 mRNA were tested by RT-PCR. Resu ltsAbB extracts markedly inhibited the proliferation of tumor cells by a dose-dependent and time-dependent manner. One of it′s anti-tumor funct ion is to block cell cycle to G 0/G 1 and lead apoptosis. Treatment with eAbB, the productions of TNF-α and IL-6 were markedly increased with dose-depende nt manner, also the expression of IL-6 mRNA was enhanced. Conclu sion The eAbB has the obvious antitumor effects. Its anti-tumor fun ction through blocking cell cycle to G 0/G 1 and leading the cell to apoptosis . Although the result of this research shows no promotional proliferation of the splenocytes of mouse, it markedly stimulates the proliferation of macrophage, n ot only enhances its phagocytosis but also improves the production of TNF-α, I L-6 and the expression of IL-6 mRNA.
