应用单细胞凝胶电泳技术检测K562细胞DNA损伤的研究

摘要 目的 用单细胞凝胶电泳技术来探讨一种能方便检测细胞DNA损伤的方法。方法 在Singh等人的方法的基础上,加入过氧化氢作为DNA致突变剂来观察K5 6 2细胞的DNA损伤。结果 在培养K5 6 2细胞的过程中,加入了不同浓度的过氧化氢,我们发现了DNA损伤与过氧化氢剂量之间存在着线性相关关系。结论这种在本实验室建立起来的用于检测细胞DNA损伤的方法易于操作,结果令人满意,并更具实用性。 Objective: To explore a method for assaying cell DNA damage by single cell gel electrophoresis (SCGE). Methods: On the basis of the research made by Singh et al., Hydrogen peroxide was added as a DNA mutagen to observe K562 cell DNA damage. Results: K562 cell strain was cultured and various concentrations of Hydrogen peroxide were added, and a linear relation between DNA damage and the dose of Hydrogen peroxide was found. Conclusion: The method for assaying cell DNA damage established in our laboratory proves to be convenient, practical and satisfactory.
