凋亡相关基因PNAS-2参与硫化砷抗APL细胞的研究 被引量:3

摘要 目的探讨凋亡相关基因PNAS2在硫化砷抗急性早幼粒细胞白血病(APL)中的作用。方法应用基因表达谱芯片检测NB4细胞在10mg/L硫化砷作用前后的基因表达改变,RT-PCR方法验证基因表达谱芯片结果和检测白血病原代细胞中PNAS2基因的表达情况。结果10mg/L硫化砷作用后的NB4细胞中,PNAS2基因表达特异性下调,且呈时间依赖性,类似变化在K562和U937细胞中未见;10mg/L硫化砷作用后2例M3患者的原代细胞PNAS2基因表达明显下调,1例M4患者原代细胞该基因表达未见明显改变。结论PNAS2基因可能是硫化砷抗APL的靶基因之一。 Objective To study the mechanism of PNAS-2 gene in acute promyelocytic leukemia(APL) cells treated by arsenic sulfide. Methods Searching for the alterably expressed genes before or after 10 mg/L sulfide by gene chip in NB4 and testing the results by RT-PCR. By the same methods we detected the expression of PNAS-2 in primary cultured cells. Results It showed in NB4 cells PNAS-2 transcripts were in lower amounts after 10 mg/L arsenic sulfide, and the effect was time-dependent, but in K562, U937 cells, the same outcome were not found. In the cells of the M 4 patient, PNAS-2 gene expression had no difference in untreated or treated with arsenic sulfide, but among M3 patients, after arsenic sulfide treated 4 h, the expression of PNAS-2 was down-regulated in 2 patients. Conclusion The gene might be the target of arsenic sulfide treated APL cells.
