致敏树突状细胞介导CTL抗骨髓瘤免疫的实验研究 被引量:1

Research of Specific Cytotoxicity T Lymphocyte Response on Myeloma Cells Induced by Myeloma Antigen-Pulsed Dendritic Cells

摘要 目的比较两种树突状细胞(骨髓瘤致敏DC和未致敏DC)诱导的细胞毒T淋巴细胞抗自身骨髓瘤的细胞免疫。方法用多发性骨髓瘤(MM)患者骨髓CD34+细胞诱生DC,将MM患者骨髓瘤冻融物冲击致敏DC。MTT法检测骨髓瘤抗原致敏和未致敏DC诱导的自身T细胞对患者骨髓瘤细胞的杀伤率。结果骨髓瘤冻融物致敏DC诱导的自身T细胞对患者骨髓瘤细胞的杀伤率远大于非致敏DC。结论患者骨髓瘤冻融物致敏的DC能有效诱导自身T细胞抗骨髓瘤免疫。 Objective To compare cell immunity of cytotoxic T lymphocyte response on myeloma cells induced by dendritic cells with or without myeloma antigen. Methods Dendritic cells (DC) were cultured by CD34 + cells from bone marrow of multiple myeloma (MM) patients. DCs were pulsed with myeloma lysates from MM patients. The killing rates of autologous T lymphocytes induced by DC pulsed with or without myeloma antigen on myeloma cells were measured by MTT method. Results Autologous T lymphocytes induced by DCs pulsed with myeloma lysates had stronger killing effect on myeloma cells than T lymphocytes induced by non myeloma antigen pulsed DC. Conclusion DCs pulsed with myeloma lysates from MM patients can effectively induce autologous T cells anti-myeloma cell immunity.
