河南省HIV-1流行毒株pol基因的分型与系统发生分析 被引量:7

Subtyping and phylogenetic analysis of pol gene sequences for HIV-1 strains isolated in Henan province

摘要 目的了解河南省艾滋病病毒1型(HIV1)流行毒株pol基因的亚型及其系统发生情况。方法采集河南省20名艾滋病病人的抗凝全血,分离血浆,用逆转录聚合酶链反应(RTPCR)扩增pol基因部分序列(PR与RT的p55区)并直接进行序列测定,登陆Web站点http://hivweb.lanl.gov确定毒株亚型。用BioEdit和DNASTAR软件的MegAlign模块进行系统发生分析,SPSS软件包对数据进行统计学分析。结果20份标本的pol基因序列系统发生分析显示均为B亚型,且具有较高的系统发生同源性,与东北地区流行毒株的相似度≥97%,PR基因离散率<0.2%,RT基因平均离散率为3.15%(0.7%~5.1%)。PR和RT基因遗传变异的差异具有显著的统计学意义(P<0.001)。结论pol基因的系统发生分析可以作为河南省HIV1毒株亚型确定的方法。河南与东北两地pol基因相似性以及PR与RT基因进化差异的意义,有待进一步研究。 Objective To undertake pol gene's subtyping and phylogenetic analysis for HIV-1 strains isolated in Henan Province.Methods Twenty plasma samples were collected from anticoagulative whole blood,for which reverse transcription-polymerase chain reaction(RT-PCR) technique was used to amplify the partial pol gene(PR and p55 of RT).The sequences' phylogenesis were analyzed by landing the websites http://hiv-web.lanl.gov,and BioEdit,DNASTAR and SPSS software.Results The phylogenetic analysis of 20 samples' partial pol sequences revealed that all of the samples were subtype B with high homology.The comparability with Dongbei's HIV-1 ≥97%;the genetic distances of PR gene were 0.2%;the average genetic distances of RT gene were 3.15%(rang,0.7-5.1%).The difference of evolution between PR and RT gene were remarkable,P<0.001.Conclusion The phylogenetic analysis of pol gene was an useful method for subtyping Henan' strains of HIV-1.The comparability of Henan and North-eastern region Dongbei's HIV-1 pol gene and the difference of evolution between PR and RT gene need further research.
