摘要
本文研究了血小板活化因子(PAF)受体拮抗剂BN52021及BN50739对内毒素所致的红细胞膜损伤的保护作用。结果表明,PAF受体拮抗剂能显著改善内毒素所致的红细胞膜磷脂降解及溶血磷脂的聚集,明显降低红细胞膜损伤过程中脂质过氧化产物的堆积及稳定膜Na ̄+-K ̄+-ATPase活力。提示PAF可能是通过活化磷脂酶A_2引起红细胞膜磷脂降解及促进氧自由基的释放而发挥作用,从而在内毒素所致细胞损伤的病理生理过程中起到重要作用。
he protective effects of platelet activating factor(PAF)receptor antagonists,BN52021 andBN50739,on the red blood cell membrane injuries induced by E. coli endotoxin administration werestudied on Wistar male rats.The results showed that BN52021 and Bn50739 significantly preventedthe membrane phospholipids strucrure from Phospholipase A2 involved degradation and theoverproduetion of lyso-PC,and significantly reduced the accumulation of ilpid peroxidation(MDA),and stabilize the Na ̄+-K ̄+-DATPase activities of red blood cell membrane. These findingssuggest that PAF may act through the membrane phospholipids degradation induced byphospholipase A2 activation and the release of oxygen free radicals. Therefore,PAF may plays acritical role in the process of red blood cell membrane damage induced by endotoxin adininistration.
