摘要
采用单克隆抗体(单抗)和WHO67/97标准血清建立ELISA法测定人血清免疫球蛋白G(IgG)亚类浓度。结果表明:本法敏感(最低可测值为2.1ng/ml,可测范围为100倍),精确(不同批测定平均变异系数≤8.06%)。所用单抗特异性强,反应性好,IgG亚类之和与常规单扩法所测总IgG值密切相关(r=0.96,P<0.01).强调将单抗包被于经BSA-戊二醛处理的微孔板上,能明显提高单抗吸附力和反应性;两种单抗混合包被可增加IgG亚类同种异型覆盖面。
ith monoclonal antibodies(McAbs )and 67/97 Standard sera, Serum concentrations of normal humanIgG Subclasses were measured by ELISA. It was emphasized that coating McAbs on the microtitre well pretreated with BSA-glutaraldehyde couldprovide the adherence and responsitivity of McAbs ,and the coating with the two mixed McAbs could enlargethe covering field of igG subclass isotypes.
