低剂量辐射联合pEgr-P16基因治疗诱导食管癌细胞凋亡的研究 被引量:2

摘要 目的:研究大剂量(2Gy)和低剂量(0.075Gy)X射线诱导pEgr P16重组质粒稳定转染的食管癌细胞株EC9706凋亡的作用。方法:将脂质体包裹的pEgr P16重组质粒,转染食管癌细胞系EC9706中,经G418筛选,获得稳定表达的细胞;采用Westernblot方法检测不同剂量X射线诱导后P16的表达;观察不同剂量X射线照射后,EC9706细胞凋亡率的变化。结果:pEgr P16重组质粒转染EC9706细胞,并获得稳定转染的细胞;2Gy和0.075GyX射线照射均可诱导P16表达增强;稳定转染的细胞经2Gy和0.075GyX射线照射,细胞凋亡率明显高于假照射(0Gy)组(P<0.05~0.01)。结论:0.075GyX射线照射可诱导稳定转染的EC9706细胞中P16表达增强,使诱导细胞凋亡率提高。 Objective:To study apoptosis induction effect of low dose and high dose X-ray irradiation on esophageal cancer cell line EC9706 transfected with pEgr-P16 plasmid.Methods:EC9706 cell line was transfected with pEgr-P16 plasmid packaged by lipsome,and steady expression cells were gotten by G418 selecting.P16 expression was detected by Western blot.EC9706 cells apoptosis was observed after different doses of X-ray irradiation.Results:pEgr-P16 plasmid was transfected into EC9706 cells and steady expression cells were gotten.High dose and low dose X-ray irradiation could enhance expression of P16.The apoptosis rates in transfected cells received high and low doses of X-ray irradiation were significantly higher than those of 0 Gy group(P<0.05~0.01).Conclusion:Low dose X-ray irradiation can enhance expression of P16 in pEgr-P16 transfected cells and induce cell apoptosis.
