摘要
目的:起源于原始粒细胞和原始单核细胞的急性白血病(acuteleukemia,AL),其临床表现和预后的具有很大的差别。研究利用蛋白质组学方法分析粒细胞来源和单核细胞来源的AL细胞差异蛋白质表达谱以了解二者差异的分子基础。方法:利用二维凝胶电泳将来源于20例粒细胞AL和10例单核细胞AL患者细胞的蛋白质进行分离,对二者的差异蛋白利用MALDI鄄TOF鄄MS生物质谱和电喷雾质谱(ESI鄄MS/MS)进行鉴定分析。结果:发现了一组粒细胞白血病和单核细胞白血病细胞的差异表达蛋白质,其中,膜磷脂A2在单核细胞白血病细胞中较粒细胞白血病细胞高表达,而上皮型脂肪酸结合蛋白在粒细胞白血病细胞中高表达。结论:为分析粒细胞AL和单核细胞AL细胞生物学差异的分子特征奠定了基础。
Objective: Acute leukemia (AL) is a heterogeneous disease with disti nct biological and prognostic groupings. AL derived from granulocytic lineage ha s different clinical characteristics from AL derived from monocytic lineage. The distinct proteins that contribute to each type were analyzed by proteomic techn ology in our study to lay the molecular basis for the different treatment reacti on and prognosis for each type, and to facilitate molecular definition of the su btypes of AL. Methods: The proteins of leukemic cells from 20 cases of granulocy tic AL patient and 10 cases of monocytic AL patients characterized by FAB classi fication were separated by two-dimensional electrophoresis(2-DE), and the differ entially-expressed protein spots between the two groups were identified by both matrix assisted laser desorption/ionization-time of flight-mass spectrometry(MAL DI-TOF-MS) and electronspray ionization(ESI)-MS/MS. Results: We found a group of the distinct expressed proteins between granulocytic AL and monocytic AL cells. Among them, annexin A2 was highly expressed in monocytic AL cells as compared t o granulocytic AL cells. Epidermal fat acid binding protein was highly expressed in granulocytic cells. Conclusion: These data have implications in molecular de finition of the different clincical characteristics between the monocytic AL and granulocytic AL.
