摘要 目的研究表皮生长因子(EGF)对神经干细胞迁移的影响.方法取孕13~14 d SD大鼠胚胎纹状体行神经干细胞培养,7 d后取悬浮神经球制成单细胞悬液再培养,EGF作用下连续培养14 d后分为实验组和对照组,继续培养,观察EGF对神经干细胞迁移的影响.结果原代培养的细胞球是神经干细胞,EGF传代培养14d时细胞球大小约100个细胞左右,实验组观察到第14d细胞球增殖变慢,细胞球长出突起与邻近的细胞球相接触,14~17 d观察到神经干细胞迁移,17 d后细胞迁移现象消失;对照组中未观察到细胞迁移.结论EGF在某一特定时间内能够诱导神经干细胞迁移. Objective To investigate the effect of epidermal growth factor (EGF) on the immigration of neural stem cells. Methods The neural precursor cells derived from striatum of 13 or 14-days-old embryos of SD rat were cultured for 7 days and then the floating neural stem cell spheres were gotten. The cells in suspension by scattered neural stem cell spheres were cultured by passage in the culture fluid with EGF for 14 days, and then were divided into experimental and control groups. The cells in the experimental groups continued to be cultured in the culture fluid with EGF, and the cells in the contro group were cultured in the cultured fluid without EGF. Results The cell spheres in the primary culture is neural stem cell spheres; A cell sphere cultured by passage for 14 days con-tained about 100 cells; In the experiment group, the prolioferaion of cell sphere became slow and the evection growing out of the cell sphere contacted with the neighbor cell sphere 14 days after the continued culture, and the neural stem cell immigrated along the evection during 14~17 days. The cell immigration was not observed in the control group. Conclusion EGF can induce the neural stem cells in vitro to immigrate in the special time.[
