摘要
体外净化除去移植物中残留的白血病细胞是提高ABMT治疗白血病效果的的关键之一。本文观察了新血卟啉光敏剂癌光啉加光照对人AML—CFU的体外净化效果。结果表明,癌光啉(10mg/L)单用对正常GM-CFU和AML-CFU集落形成均无明显影响,照光后集落存活率随照光时间延长而下降,照光4分钟CFU—GM和AML—CFU存活率分别为对照的23.374±25.66%和1.63±3.24%,照光6分钟,12例AML中无一例生长,此时CFU—GM存活率为3.51±3.43%。说明癌光啉—光照在体外对白血病细胞确有净化作用,可用于临床ABMT。另外,不同个体对癌光啉光照的敏感性相差较大,提示临床进行体外净化时应先行敏感性试验,以确定最适照光时间。
One of the important problems of autologous bone marrow transplantation for the treatment of leukemia is to purge the residual leukemic cells from the autologus marrow grafts in vitro.This paper reported the selective killing effect of photocarcinorin(PsD-007),a new photosensitve agent from hematoporphyrin,plus light irrdiation on human leukemit progenitors of acute myelobastic leukemia(AML-CFU)and normal granulocytic and macrophagocytic progenitors (GM-CFU).It was observed that photocarcinorin(10mg/L)alone did not affect the viability of both AML-CFU and GM-CFU.After daylight flurescence irradiation,the colony recovery rate of AML-CFU and GM-CFU decreased as irradiation time went,with the former decreased more signiticantly.After 4 minutes of irradiation,the survival rate of GM-CFU and AML-CFU were 23.37±25.66% and 1.63±3.24%,respectively.when no AML-CFU(n=12)appeared after 6 minutes of irrradiation,3.51±3.43% of CFU-GM remained survival.The result suggesed that photocarcinorin and light could eliminate leukemic progenitors,and might be used in clinical ABMT.We also found that irradiation time should be adjusted individually before purging,for the sensitivities of GM—CFU and AML—CFU from different individuals to photosensitzation were quite different.
