GH对创伤与感染SD大鼠IGF-1水平及营养代谢的影响

摘要 目的 探讨创伤与感染及外源性生长激素对SD雄性大鼠血清胰岛素样生长因子 1(IGF 1)及营养代谢的影响。 方法 SD雄性大鼠 6 0只 ,随机分成生长激素治疗组和对照组 (各 30只 ) ,复制成腹腔感染动物模型 ,治疗组术后第 1天开始给予GH 0 .5U·kg 1·d 1连续 7天 ,对照组给予相同体积的生理盐水。分别测定术前及术后第 1、4、7天血清GH、IGF 1、总蛋白、白蛋白、前白蛋白、转铁蛋白及血糖变化。 结果 术后两组SD大鼠均出现血清IGF 1、总蛋白、白蛋白、前白蛋白及转铁蛋白降低和血糖、GH明显提高 ,治疗组大鼠的GH及血糖明显高于对照组 ,而两组之间IGF 1及血清蛋白水平于术后第 1、4天差别无显著性。术后第 7天治疗组大鼠IGF 1、前白蛋白及转铁蛋白水平高于对照组。 结论 外源性生长激素能够明显提高血清GH及血糖水平 ,但在术后早期对IGF 1及血清蛋白却未产生显著性影响 ,故认为应在机体度过急性应激期后方适合使用外源性生长激素。 Objective (1) To investigate the changes of metabolism and serum insulin like growth factor 1(IGF 1)level in male Sprague Dawley rats after trauma and infection;(2)To investigate the influence of the administration of exogenous growth hormone(GH) on the IGF 1 level and metabolism. Methods Sixty male SD rats(weight 290~310g)were randomly divided into GH group and controlled group (n=30).All rates were replicated into abdominal infection models by operation.The GH group were given GH(0.5Ukg 1day 1)for 7 days and the controlled group received saline.Blood samples were collected and analyzed for GH,IGF 1,total protein,albumin,prealbumin,transferrin and glucose concentrations a day before and 1,4,7 days after operation. Results The concentrations of IGF 1,total protein,albumin,prealbumin and transferrin decreased to some degree after operative trauma and infection,but the concentration of glucose and GH was elevated.The administration of exogenous GH promote hyperglycemia and elevate GH level,but the IGF 1,total protein,albumin,prealbumin and transferrin levels are not sygnificantly elevated 1 day and 4 days after operation Conclusions Because of the presence of GH resistance there is no indication of administration of exogenous GH early after trauma and infection.
