摘要
本文建立了用GC-MS测定血浆中可待因、吗啡的分析方法。本方法是以烯丙吗啡为内标物,采用液相提取,衍生化,进行定量测定。可待因在1-100 ng·ml^(-1)浓度范围,线性关系良好,灵敏度为1 ng·ml^(-1);吗啡在2-100 ng·ml^(-1)浓度范围,线性关系良好,灵敏度为2 ng·ml^(-1)。应用本法对两例正常人po 30 mg磷酸可待因进行药代动力学研究。
An analytical procedure for simultaneous determination of codeine and morphine in plasma was described in this paper. The detection of codeine and morphine was based on liquid-liquid extraction and derivatization to the acetylated compounds. The acetylated codeine and morphine were separated by capillary gas chromatography and iden- tified mass spectrometrically by selected ion monitoring(SIM). Quantitative determination was carried out by SIM using nalorphine as internal standard. Calibration curves in the range 1-100 ng . ml^(-1) for codeine and 2-100 ng . ml ^(-1) for morphine were linear. The lower limits of detection for codeine and morphine were 1 ng . ml^(-1) and 2 ng . ml(-1), re- spectively.
