摘要
目的测定脂肪乳剂Intralipid在体外是否影响内毒素和细胞因子所致的巨噬细胞的一氧化氮自由基的产生。方法采用腹腔灌洗收集20头SD大鼠腹腔巨噬细胞,在体外置37℃5%CO2培养箱中孵育,应用大肠杆菌脂多糖和γ-干扰素刺激巨噬细胞。实验分单纯巨噬细胞组和Intralipid组,采用Griess反应测定NO的产生量。结果经LPS、IFN-γ刺激后,巨噬细胞的NO产生量明显高于静止组(P<0.05)脂肪乳剂可明显降低刺激组和静止且巨噬细胞的NO产生(P<0.05)。结论Intralipid词在体外可抑制巨噬细胞介导的一氧化氮自由基的产生。
Objective To study the effect of Intralipid on the macrophages-induced release of nitric oxide radicals (NO) in vitro.Methods Peritoneal macrophages of SD rats were incubated either with purified cell envelope lipopolysaccharides (LPS) or interferon-γ(IFN-γ) with or without Intralipid.The release of NO was measured by the Greiss reaction.Results The results showed that the incubation of macrophages with LPS or IFN-γ increased their oxidative metabolism with increased release of NO.However,when the macrophages were incubated with LPS or IFN-γ together with Intralipid, the preduction of NO was significantly decreased.Conclusions Intralipid might substitute for plasma high density lipoprotein, which are known to inhibit the LPS effect on the macrophages in the acute stage of an infection with Gram-negtive bacterial.
