摘要
海洋贝类中多糖和蛋白含量很高,不易获得高纯度、完整的基因组DNA,且海洋贝类很易死亡腐败,将引起DNA的降解。而获得高纯度的完整DNA是进行DNA指纹图谱、RAPD分析和DNA测序等技术操作的前提。SDS方法可有效地去除多糖、蛋白、RNA、酚等杂质,且可从无水乙醇固定的标本中提取高质量DNA。
Seashells have low contents of polysaccharide and protein, which makes it difficult to extract high- purity unbroken genome DNA. Seashells are also easy to die and decay and that will cause DNA to degrade. The extraction of high- purity unbroken genome DNA, however, is the precondition of technical operations, including DNA fingerprint, RAPD analysis and DNA sequencing. The use of SDS method can effectively leach polysaccharide, protein, RNA, hydroxybenzene and other impurities and also extract high- purity unbroken genome DNA from fixed specimens with absolute ethyl alcohol.
