前荷叶碱对人脐静脉内皮细胞一氧化氮及一氧化氮合酶的影响 被引量:7

The Effects of Pronuciferine on NO and NOS Production in Cultured Human Umbilical Vein Endothelium Cells

摘要 目的观察前荷叶碱(Pronuciferine)对人脐静脉内皮细胞(HUVECs)分泌一氧化氮(NO)和一氧化氮合酶(NOS)的影响,探讨其对HUVECs血管活性分子分泌的调节作用。方法从莲子心中提取前荷叶碱,体外培养HUVECs,用不同浓度(10μmol/L、1μmol/L、0.1μmol/L、0.01μmol/L)的前荷叶碱分别作用于HUVECs,观察细胞形态,MTT法观察前荷叶碱对HUVECs活性的影响,比色法测定各组的NO、总NOS(tNOS)、诱导型NOS(iNOS)。结果和对照组比较,前荷叶碱对内皮细胞形态和活性无明显影响,能增加HUVECs释放NO,及tNOS生成(p<0.01),但对iNOS影响不大。结论前荷叶碱可能通过增加tNOS生成而提高HUVECs释放NO,从而保护内皮功能。 Objective: To investigate the effects of Pronuciferine on nitric oxide(NO), total nitric oxide synthase(tNOS) and inducible nitric oxide synthase(iNOS) production in cultured Human Umbilical Vein Endothelium Cells(HUVECs). Methods: Pronuciferine was extracted from plumula nelumbinis. The experiments were performed in HUVECs in vitro. Cell-morphology was observed by light microscope. Cells viability was assessed by MTT assay. NO, tNOS and iNOS were measured by Colorimetry. Results: Cell-morphology and viability weren’t affected by Pronuciferine. Pronuciferine significantly increased the level of NO and the activity of tNOS, but had no effect on the activity of iNOS. Conclusion: Pronuciferine increases the level of NO by the enhancement of the activity of tNOS. It may have the protection on endothelial function.
