荧光定量PCR技术检测乳腺癌患者外周血CEA、CK19 mRNA表达的意义 被引量:1

摘要 目的探讨荧光定量PCR(FQPCR)检测外周血癌胚抗原(CEA,carcinoembryonicantigen)、细胞角蛋白19(CK19,cytokeratin19)mRNA的表达在乳腺癌外周血微转移诊断中的应用。方法采用FQPCR法,并以βactin为内对照,测定19例乳腺癌组织中特异性mRNA表达情况,以及测定28例健康女性体检者、11例良性乳腺疾病患者和51例乳腺癌患者的外周血中CEA、CK19mRNA的表达。结果19例乳腺癌组织中CEAmRNA的阳性率为78.9%(15/19),CK19mRNA的表达阳性率为100%(19/19)。本研究应用FQPCR检测外周血CEA、CK19mRNA的表达来检测乳腺癌外周血微转移,以上有一项为阳性即判定为转移。正常对照组和良性乳腺疾病组CEA、CK19mRNA的表达均为阴性,乳腺癌组显著高于前2组。结论FQPCR技术是高度灵敏、高度特异的快速定量检测CEA、CK19mRNA的方法,可有助于乳腺癌的诊断和评估预后。 Objective To evaluate CEA,CK19 mRNA detected by fluorescent quantitative polymerase chain reaction(FQ-PCR)as tumor markers for micrometastatic carcinoma cells in the peripheral blood of patients with breast cancer.Methods β-actin was used as internal control.Total RNA extracted from 19 breast carcinoma tissue was detected by FQ-PCR.Clinical samples of 28 healthy women,11 patients with benign breast disease and 51 patients with malign breast disease were detected by FQ-PCR.Results In 19 breast carcinoma tissue,the CEA mRNA detection rate were 78.9%(15/19),the CK19 mRNA detection rate were 100%(19/19).We examined CEA,CK19 mRNA expression by FQ-PCR in peripheral blood samples of patients with breast neoplasms and healthy controls.We determined the sample was positive if either CEA mRNA or CK19 mRNA was detected in peripheral blood samples.Blood samples were positive in patients with breast carcinomas.None of the patients with benign breast neoplasms and healthy controls was positive.Conclusion The FQ-PCR technique was a rapid quantitative method with high sensitivity and speciality in detecting micrometastases of breast cancer.It may be a efficient method to guide the diagnosis of breast cancer and estimate the prognosis.
