摘要
目的:探讨丝裂霉素(MMC)对小鼠淋巴瘤EL- 4细胞P2 1蛋白表达的影响。方法:采用免疫荧光染色及流式细胞术检测P2 1蛋白表达的时程和量效改变。结果:时程实验研究证实,给予2 m g·L- 1 MMC处理后,EL- 4细胞中P2 1蛋白表达在2~4 8h明显高于对照组(P<0 .0 5 ,P<0 .0 1)。量效实验结果证实,给予1、2和4 mg·L- 1 MMC处理后2 4 h,P2 1蛋白表达与对照组相比明显增高(P<0 .0 1)。结论:MMC能诱导P2 1蛋白表达增高,并存在时间和剂量依赖关系。
Objective To explore the effects of mitomycin (MMC) on the expressions of P21 protein in EL-4 mouse lymphoma cells. Methods Flow cytometry and immunofluorescence staining were used to measure P21 protein expressions in changes with time and doses. Results In time-course experiments, it was demonstrated that P21 protein levels were markedly increased at 2-48 h after treatment with 2 mg·L -1 MMC in EL-4 cells compared with control group (P<0.01, P<0.05). In dose-effect experiments, it was showed that P21 protein expressions were increased significantly 24 h after treatment with 1,2,and 4 mg·L -1 MMC in EL-4 cells compared with control group (P<0.01). Conclusion The P21 protein expression could be increased by MMC in time- and dose-dependent manners.
