己酮可可碱对血管成形术后管腔狭窄的影响 被引量:3

The Influence of Pentoxifylline on the Restenosis after Angioplasty

摘要 目的 探讨己酮可可碱(PTX)对实验性兔髂动脉球囊损伤术后修复过程中血管狭窄的干预作用。方法 建立兔髂动脉球囊损伤后狭窄模型5 0只,随机分成:(1)单纯球囊损伤组(n =2 0 ) ;(2 )PTX组(n =2 0 ) ;(3)假手术组(n =10 ) ,于血管成形术后4周处死各组动物,取目标血管段计算机图象分析检测血管管腔面积、新生内膜面积、胶原特殊染色平均灰度值。结果 用药组与空白对照组的内膜胶原染色平均灰度分别为18.6±1.5 6与2 9.3±2 .71,P <0 .0 1;两组血管管腔面积差异显著(1 18±0 .15vs .0 .84±0 .11;P <0 0 1) ;用药组较空白对照组的新生内膜面积减少(0 .6 7±0 .0 7vs.0 .90±0 .13:P <0 .0 1)。结论 PTX能通过显著抑制内膜胶原的增生,提高管腔面积,从而降低了血管损伤后狭窄的程度。 Objective To study the effect of pentoxifylline(PTX) on restenosis duffng the vessel repairing after balloon overstrech injury of rabbit iliac arteries.Methods Fifty adult male rabbits were divided into three groups at random:(1) balloon group(n=20):received balloon injury only;(2)PTX group(n=20):imraperitoneal irrigated of PTX from 1 day beforeinjury at a dose of 42mg/kg till the animals being harvested at week 4;(3)sham group(n=10).The illac arteries were analyzed by cross-sectional morphometry and the lumen area(LA) and neointimal area (NA) calculated.We also examined the mean greym of collagen within the neointima by Van Gieson stain to look into collagen producing.Result The collagen greym of neointima in PTX group and in balloon group was prominently difierent (18.6±1.56 vs.29.3±2.71;P<0.01).The LA of PTX group was 1.18±0.15mm^2 and that of balloon group was 0.84±0.11mm^2 (P<0.01).The NA of PTX group was significantly lower than balloon group (0.67±0.07mm^2 vs. 0.90±0.13mm^2;P<0.01).Conclusion PTX may pretect the artery from stenosis by weakening collagen production and then improving the lumen area after balloon injury.
