摘要
目的 观察老年人血小板内钙浓度([Ca2 + ]i)的变化。方法 以Fura/AM为荧光指示剂,采用荧光分光光度计测定分离的老年及成年人血小板内[Ca2 + ]i。结果 在细胞外钙为1mmol·L-1的情况下,静息状态老年人血小板[Ca2 + ]i及对高钾除极化的反应均明显低于青年对照组(P <0 .0 1) ,高钾使血小板内[Ca2 + ]i增加的绝对值在老年人与青年人间无明显差异(P>0 .0 5 )。结论 老年人血小板[Ca2 + ]i降低,但在相同情况下老年人血小板胞内钙超载更加明显,从而较青年人更易发生细胞的损害。
Objective To observe the change of intraplateletic free calcium concentration (i) of aging persons.Method Fluorescence spectrophotometer with fura-2 as a calcium fluorant was used to measure i of dissociated platelets from aging person and adults.Result In the presence of extracellular Ca^(2+) 1mmol·L^(-1) resting i and the response of rise in i to KCl in platelets from aged persons were all significantly reduced than those from young persons,but it significantly enhanced the increase of i induced by different concentration of KCl in both group,and the enhancement was not significantly different between the aging and the young(P>0.05).Conclusion Resting platelet i in aging persons is reduced but,plateleti in aging persons overburdened more significantly than that in the young under same circumstances,and then the cells of aging persons more easily get damaged.
