脾虚早期与脾虚期大鼠组织和血浆中SS、CCK、MOT含量变化的探讨 被引量:16

摘要 目的 探讨胃肠激素在脾虚证大鼠体内的动态变化规律,进一步揭示脾虚证的本质。方法 随机将SD大鼠分为5组,采用利血平[0 .3mg/ (kg .d) ]股四头肌注射,作用于大鼠7d和14d分别造成脾虚早期和脾虚期模型,用四君子汤做反证,六味地黄汤作阳性对照。用放射免疫法(RIA)检测不同脾虚造模组及对照组大鼠血浆,胃窦、十二指肠、空肠及下丘脑组织中生长抑素(SS)、胆囊收缩素(CCK)和胃动素(MOT)的含量变化。结果 与正常对照组比较,脾虚早期( 7d) :SS在血浆、下丘脑中含量升高(P <0 .0 1或P <0 .0 5 ) ,外周组织中含量下降(P <0 .0 1) ;CCK和MOT在组织和血浆中含量均升高(P <0 .0 1或P <0 .0 5 ) ;脾虚期( 14d) :SS在血浆、下丘脑中含量下降,外周组织中含量升高(P <0 .0 1) ;MOT在组织和血浆中含量均下降;CCK在下丘脑及胃窦组织中含量升高(P <0 .0 1) ,在血浆和肠道组织中含量下降(P <0 .0 1)。四君子汤预防性治疗的大鼠体内胃肠激素含量变化均无显著差异;六味地黄汤组胃肠激素变化有显著差异。结论 脾虚证存在多种胃肠激素的异常,且随不同脾虚时期发展呈动态变化,四君子汤对此有纠正作用。 Objective To study the changes of gastrointestinal hormones in rats with spleen asthenia syndrome according to traditional Chinese medicine and the basic mechanism of spleen hypofunction.Methods The rat models with spleen hypofunction were developed by injecting reserpine [0.3 mg/(kg·d)] into the quadriceps femoris of Shanghai SD rats for seven days or fourteen days.Si Junzi Tang (Decoction of Four Noble Drugs) and Liuwei Dihuang Tang Decoction of Six (Drugs Including Rehmannia) were used for treatment.RIA was used to detect the levels of somatostatin (SS),cholecystokinin (CCK),and motilin (MOT) in tissues of gastric antrum, duodenum jejunum and hypothalamus or plasma.Results Compared with normal rats,in the rats with spleen hypofunction at early stage,SS levels were higher in plasma and hypothalamus(P<0.01 or P<0.05),and lower in peripheral tissues(P<0.01);CCK and MOT levels were higher in tissues and plasma(P<0.01 or P<0.05).In late stage of spleen hypofunction,SS levels were lower in plasma and hypothalamus,and higher in peripheral tissues (P<0.01);MOT levels were lower in both plasma and hypothalamus;CCK levels were higher in hypothalamus and gastric antrum (P< 0.01 ) and lower in plasma and intestinal tissues (P<0.01).Conclusions Rat models with spleen hypofunction were successfully developed.Abnormities of gastroinfestinal hormones exist in muttiple organs and vary with different stages of spleen hypofunction.Si Junzi Tang can correct the syndrome but Liuwei Dihuang Tang cannot.
