摘要
目的:建立高效液相色谱法(HPLC法)测定鼠脑组织中氨甲喋呤(MTX)浓度的方法。方法:脑组织样加入高氯酸,残渣经流动相溶解后测定,检测波长为306nm。结果:相关系数r=0.9997;线性范围:10.0~0.078125μg.ml-1;最低定量浓度为0.078125μg.ml-1;日内RSD≤7.00%,日间RSD≤6.44%,萃取回收率RSD为3.28%~5.48%。结论:本方法简便、可靠,灵敏度高,可用于氨甲喋呤在大鼠脑组织内的检测和药代动力学研究。
Objective:To establish the valid detection method with the HPLC system for the determination of concentration of MTX in rat brain.Methods:MTX was extracted from brain tissue get in with HClO 4 and soluted by a mobile phase and then detected at 306nm. Results:The correlation coefficients was 0.9997.The calibration curve was linear over the range of 10.0~0.078μg.ml -1,and the limit of detection was 0.078μg.ml -1.The average relative deviations of within day and between day variation were 2.82%~7.00% and 4.19~6.44% respectively.The average relative deviations of recovery of MTX in sample was 3.28%~5.48%.Conclutions:The method was simple,sensitive and accrate.It was shown to be suitable for monitoring and pharmacokinetic studies of MTX in rat brain.
