摘要
目的:了解局灶性脑缺血再灌注后Nestin的动态变化,以及三七三醇皂苷对其表达的影响。方法:采用改良的线栓法制备大脑中动脉阻塞2h、不同再灌注时间段(3h、6h、12h、24h、48h、72h、7d、14d)的大鼠短暂局灶性脑缺血模型,应用免疫组织化学技术检测巢蛋白的表达。结果:脑缺血再灌注后24h,PTS(三七三醇皂苷)组缺血侧Nestin即有增加,并持续至7d,较之盐水对照组差异显著,至再灌注14dNestin表达已明显回落。结论:PTS可使缺血再灌注大鼠的Nestin表达上调,这可能是其发挥脑保护作用的机理之一。
Objective:To study the expression of Nestin and the effect of PTS on it following focal ischemia-reperfusion in the adult rat brain.Metheods:The model of acute reperfusion injury after cerebral ischemia in rats was made by middle cerebral artery occlusion.All animals were allowed 2h ischemia and sacrificed after different durations of reperfusion (3h,6h,24h,48h,72h,7d,14d).The expression of Nestin was detected with an immunohistochemical approach.Results:The number of Nestin-immunoreactive cells at 24h after reperfusion in ischemic side in PTS group started to increase markedly and represent significant difference compared with the N.S group,peaked on 7d and then decreased observably on 14 d after reperfusion.Conclusion:The study implies PTS could up-regulate the expression of Nestin,and it might be the one of protective mechanisms of PTS against ischemia-reperfusion injury.
