链脲佐菌素诱导大鼠糖尿病肾病模型的方法学探讨 被引量:51

摘要 目的:对比观察不同方法对链脲佐菌素(STZ)诱导速发型糖尿病肾病(DN)模型的影响。方法wis tar和SD大鼠,空腹或非空腹状态下,STZ一次腹腔注射,72h后监测血糖、尿蛋白以及一般项目,同时行肾胰病理检查。结果:空腹wistar鼠成模率62.5%,SD鼠为87.5%,非空腹组成模率是10%,未成模鼠小剂量空腹追加STZ无一例成模。成模鼠肾胰显示特征性改变。不同时点血糖值差异大,第二周平均尿蛋白排泄即达33.48mg/24h。结论:非空腹鼠成模率极低,SD鼠成模可能优于wistar鼠,成模前血糖至少连续三天同时点监测,对早期DN的观察最好始于成模后第一周。 Objective:To compare observation of diabetic nephropathy(DN)induced by streptozotocin(STZ)in the different stock rats and condition.Methods:Under the empty or full stomach,DN model in Wistar or Sprague-dawley(SD)rats was induced with STZ once intraperitoneal injection.The changes of blood sugar,proteinuria,and general index were monitored.The tissue sections of renal and pancreas were stained by HE.Results:The rate of developed model in Wistar and SD rats of the empty stomach was 62.5%,87.5%,respectively,While Wistar rats of the full stomach was 10%.The value of the blood sugar was greatly fluctuant after STZ injection.In developed model,urine protein count exceed 30mg/24h in 2 nd week,and pathological changes occured in kidney and pancreas.Failed rats were not duplicated by the low dose STZ reinjection.Conclusion:SD rats may be superior to Wistar rats in ND model induced by STZ, but the rate of developed ND in rats of the full stomach is extremely low. It is essential that 3 day value of the blood sugar is observed continually and changes of early DN are studied in 1 st week in developed model.
