摘要
从地衣芽孢杆菌BL1中提取α-乙酰乳酸脱羧酶,为提高酶的得率,探讨了破壁方法、硫酸铵盐析提取粗酶等的条件。结果表明:菌体用超声波破壁、在频率25KHz下每次辐射2s,间隔2s,总辐射时间4.6s,输出功率500W,细胞浓度2%,破碎产酶量最高,60%~70%饱和度硫酸铵盐析提取粗酶效果较好。
α-acetolactate decarboxylase was extracted from Bacillus licheniformis BL1.In order to get more ALDC, the extracting process at different steps was examined, such as breaking cell wall and removing contaminating proteins by ammonium sulfate precipitation. The conclusions was made: It had a better results when use ultrasonic wave frequency 25KHz, ultrasonic power 500W, cell density 2%, radiate 2 second each time, total radiate 4.6 seconds, 2 second interval to break cell wall, and 60%~70% ammonium sulfate precipitation to remove contaminating proteins and extract crude enzyme products.
