摘要
目的:探讨RU486对卵巢过度刺激综合征(OHSS)模型大鼠血管内皮生长因子(VEGF)产生的作用。方法:30只22日龄雌性大鼠,从22日龄起每天皮下注射10IUPMSG连续4d,26日龄时皮下注射100IUhCG,建立OHSS模型。在27日龄将上述大鼠随机分成5组,每组6只。其中4组(A-D组)为实验组,分别皮下注射不同剂量(1mg/kg、5mg/kg、10mg/kg、20mg/kg)的RU486;另一组为对照组不给药。RU486注射48h后,采用酶联免疫吸附试验方法测定血清和腹腔冲洗液中VEGF水平;采用免疫组织化学法和逆转录聚合酶链反应技术检测卵巢组织VEGF蛋白质和mRNA的表达。结果:B组和C组的血清VEGF水平分别为55.00±14.13pg/ml和49.67±17.44pg/ml,均显著低于对照组(76.17±18.19pg/ml)(P<0.05)。同样,B组和C组的腹腔VEGF水平分别为10.43±6.80pg/ml、10.30±5.82pg/ml,也均显著低于对照组(17.12±1.71pg/ml)(P<0.05)。B组和C组的卵巢组织VEGF蛋白质和mRNA的表达显著低于对照组(P<0.05)。结论:适当剂量的RU486能够降低OHSS模型大鼠VEGF的产生。
Objective: To explore the effect of RU486 on the production of vascular endothelial growth factor in the ovarian hyperstimulation syndrome model rats. Methods: A total of 30 immature female rats aged 22 days accepted s.c. injection daily with 10 IU pregnant mare serum gonadotropin (PMSG) for 4 consecutive days from the d 22 to d 25. On d 26, 100 IU human chorionic gonadotropin (hCG) was injected to produce OHSS model. On d 27, 30 rats were randomly divided into five groups. Each group contained 6 rats. The rats in group A to group D were given the dosage of RU486 with 1 mg/kg, 5 mg/kg, 10 mg/kg, 20 mg/kg, respectively and the rest group was the control. Forty-eight hours after RU486 treatment, the VEGF levels in serum and peritoneal fluid were measured by enzyme-labeled immunosorbent assay. With immunohistochemistry and reverse transcription poly- merase chain reaction, expressions of VEGF proteins and their mRNA in the ovarian tissue of each group were detected. Results: The serum VEGF levels of group C and group D were 55.00 ± 14.13 pg/ml and 49.67 ± 17.44 pg/ml respectively, which were significantly lower than that of control group(76.17±18.19 pg/ml) ( P<0.05, P<0.05, respectively). Meanwhile, the peritoneal VEGF levels of group C and group D were 10.43±6.80 pg/ml and 10.30 ±5.82 pg/ml, which were also significantly lower than that of control group (17.12±1.71 pg/ml) (P<0.05, P<0.05, respectively). The expressions of VEGF proteins and their mRNA in ovarian tissue of group Ⅲ and group Ⅳ were significantly lower than that of control group. Conclusion: Optimal dosage of RU486 could reduce the production of VEGF level in OHSS model rats.
