芪丹通脉片对内皮细胞抗血栓形成作用的影响 被引量:13

摘要 目的: 研究益气活血化瘀中药复方制剂芪丹通脉片(qidantongmaitablet,QDTMT)对血管内皮细胞(vasculaen dothelialcells,VEC)抗血栓形成作用的影响,进一步探讨益气活血化瘀药物的作用机制.方法: 48只SD大鼠随机分为空白对照组(Control)、模型组(Model)、QDTMT高剂量组(QDT -MTH)、QDTMT中剂量组(QDT-MTM)、QDTMT低剂量组(QDT MTL)和阳性对照组(Herbesser),用QDTMT(或生理盐水)给SD大鼠连续ig14d,第15日起注射大剂量肾上腺素,复制血管内皮细胞损伤模型,全自动血液凝固法检测抗凝血酶-Ⅲ(AT- Ⅲ)和血管性血友病因子(vWF)的活性,放免法检测6-酮-前列环素-F1α(6- Keto- PGF1α)和血栓素B2 (TXB2 )的含量.结果: 益气活血化瘀中药复方制剂QDTMT能显著降低血管内皮细胞损伤大鼠血浆vWF活性和TXB2 的水平,显著升高AT Ⅲ活性, 6- Keto -PGF1α水平和6- Keto -PGF1α/TXB2 比值,与模型组相比有显著差异(P<0 .01),且QDTMTH明显优于QDTMTM和Herbesser.结论: QDTMT对内皮细胞损伤具有保护作用,增强血管内皮细胞抗血栓功能和减低止血功能,这可能是QDTMT益气活血化瘀的作用机制之一. AIM: To study the effects of QiDanTongMai tablet (QDTMT),a compound preparation made up of pure traditional Chinese herbs,on the anti-thrombosis function of endothelial cells in rats with vascular endothelial injury,and to further explore the mechanism of reinforcing qi and activating blood circulation to remove blood stasis.METHODS: Forty-eight SD rats were divided randomly into Control,Model,QDTMT high dosage (QDTMT_H) and moderate dosage(QDTMT_M),low dosage(QDTMT_L)and herbesser (Herbesser) groups.The suspension of QDTMT was given to SD rats by gastrotube for 2 weeks and the model of vascular endothelial was established by injecting subcutaneously a high dose of adrenaline solution to the SD rats on the 15th day.The activity of AT-Ⅲ and vWF and the concentration of 6-Keto-PGF1a and TXB_2 were determined by automatic blood coagulation and radioimmunoassay respectively.RESULTS: Compared to those of the model group,the activity of AT-Ⅲ and the concentration of 6-Keto-PGF1a and the ratio of 6-Keto-PGF1a/TXB_2 increased.The activity of vWF and the concentration of TXB_2 were lowered significantly in the group of QDTMT_H and QDTMT_M and the effects of QDTMT_H were obviously better than those of QDTMT_M and Herbesser.CONCLUSION: QDTMT can prevent vascular endothelium from injury, increase the anti-thrombosis function and decrease the hemostasis function of endothelial cells,which may in part be responsible for QDTMT in reinforcing qi and activating blood circulation to remove blood stasis.
