摘要
目的: 研究中药“十八反”中甘遂与甘草配伍对大鼠肝脏CYP3A2在mRNA水平、蛋白表达及酶活性方面的调控作用.方法: RT PCR检测大鼠肝脏CYP3A2mRNA水平;WesternBlotting检测大鼠肝脏CYP3A2蛋白表达;通过红霉素N 去甲基反应评价CYP3A2酶活性.结果: 在mRNA水平、蛋白表达、酶活性方面,甘遂单用组均高于其他组.结论: 甘遂、甘草可能存在基于药物代谢酶机制的药物间相互作用.
AIM: To study the effects of Euphorbia coadministration with Glycyrrhiza on the mRNA,protein expression and activity of cytochrome P450 3A2 (CYP3A2) in rats.METHODS: The expression of CYP3A2 mRNA was detected by semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) and the expression of CYP3A2 protein was detected by Western Blotting.The activity of CYP3A2 was analyzed by erythromycin demethylation method.RESULTS: The expression of CYP3A2 mRNA and CYP3A2 protein,and the activity of CYP3A2 were higher in Euphorbia group than those in the other groups.CONCLUSION: Euphorbia coadministration with Glycyrrhiza may have some interaction based on CYP3A2.
