摘要 目的研究血管外膜成纤维细胞(AF)能否产生结缔组织生长因子(CTGF),以及血管紧张素Ⅱ(AngⅡ)对CTGF的影响。方法组织贴片法培养WKY大鼠主动脉AF,经AngⅡ(10-7~10-9mol/L)处理,应用免疫细胞化学和Westernblot的方法,观察CTGF的表达,及AT1受体阻断剂氯沙坦和AT2型受体阻断剂PD123319干预后的CTGF表达。结果免疫细胞化学显示AF表达CTGF,AngⅡ可以诱导AF以及其上清液中CTGF表达增加。Westernblot显示AngⅡ诱导CTGF表达呈剂量和时间依赖性。10-7mol/LAngⅡ刺激24h诱导AF产生CTGF作用最强。AngⅡ诱导CTGF表达增加作用可以被氯沙坦阻断,而不受PD123319影响。结论AngⅡ通过AT1受体诱导CTGF合成。 ObjectiveTo test the hypothesis that angiotensin Ⅱ(Ang Ⅱ) can induce the expression of connective tissue growth factor (CTGF) in vascular adventitia fibrobasts(AF). Methods The vascular AF isolated from WKY was cultured by the explant method and identified immunohistochemically. The effect of Ang Ⅱ on CTGF protein expression was assessed by immunohistochemical and Western blot method. We also studied the effects of AT 1 receptor antagonist losartan and AT2 antagonist PD123319 on CTGF protein expression. Results Ang Ⅱ increasedCTGF expression. Ang Ⅱ in a time and dose dependent manner with a maximal effect after 24 h and a dose of 10~ -7mol/L.These effects were blocked by pre-incubation with the AT1 antagonist losartan, but not AT2 antagonist PD123319.Conclusion Our results show that Ang Ⅱ increases CTGF in adventitia fibroblasts via AT 1 receptor.
