摘要
对大葱成熟胚的愈伤组织诱导及分化过程进行了观察,并对再生过程中内源激素水平进行了测定。研究结果表明:大葱愈伤组织诱导过程中,ZR在3d时含量达峰值,可能是导致6d时增长率最高的主要原因,ABA在此过程中可能为负调控因子。GA3在愈伤组织分化过程中的含量明显高于继代过程,说明其对大葱愈伤组织的分化有重要作用。适当高的6BA与NAA比值有利于芽的分化,但是细胞分裂素类浓度过高时则会对不定芽造成伤害。
The callus induction and plant regeneration from Allium fistulosum L.mature embryos culture were studied in A. fistulosum L. and the endogenous hormone changes in this course were analyzed. The callus growth rate reached the highest point at the 6th day after culturing, which might be caused by the maximum concentration of ZR at the 3rd day. ABA was a negative regulation factor in the callus induction. The concentration of GA_3 during callus induction was much higher than that in the multiplication course, which indicated GA_3 was very important in the differentiation period. The appropriate proportion between 6-BA and NAA was beneficial to the shoot differentiation of A. fistulosum L., but there was side effect to shoot regeneration if CTK concentration was too high.
