摘要
目的 探讨局部运用阿仑磷酸钠对抑制破骨细胞性骨吸收的影响,为临床上人工关节的松动提供预防和治疗的方法。方法 用20只大耳白兔建立骨质疏松模型后,在模型兔右侧股骨中上段截骨,用仿锤形不锈钢假体连接断端,一组用阿仑磷酸钠加骨水泥(PMMA)固定假体,对照组为单用PMMA固定。8周后处死全部实验兔,取出实验股骨,作生物力学实验和病理形态学检查,进行图像分析和统计学处理。结果 在拔出实验和扭转实验,阿仑磷酸钠组均显著优于对照组(P<0.01)。病理组织形态学检查阿仑磷酸钠组骨小梁体积比、厚度和数目高于对照组(P<0.01或P<0.05)。结论局部运用阿仑磷酸钠对人工假体无菌性松动有明显抑制作用,增加了人工假体的稳定性。
Objective To investigate the practical effect of the local application of alendronate on inhibiting osteolysis caused by osteoclast and provide the clinical methed of prevention and treatment on aseptic loosening of prosthesis.Methods Animal osteoporosis models were established in 20 rabbits, After osteotomy in right femur were operated, the stainless steel implants were implanted. 20 model rabbits were divided into 2 groups, with one group receiving alendronate PMMA bond, and the other group receiving PMMA bond as control. 20 rabbits were sacrificed after 8 weeks of implantation, Specimens of right femur were sectioned to be studied by biomechanical test and histomorphometry study. Results The pull-out strength and torsion moment of alendronate-PMMA group were significantly higher than that of control group(P<0.01). trabcular area percent, the thickness and the number of trabeculae were higher than that of control group(P<0.01 or P<0.05). Conclusion The local application of alendronate can inhibit aseptic loosening of prosthesis potently as a potent inhibitor of bone resorption and increase the stabilities of prosthesis in rabbit.
