脂多糖和细胞因子刺激肾小球系膜细胞合成PAF的研究 被引量:1

摘要 本研究首先建立了血小板活化因子(PAF)生物活性检测法,并测定了体外培养的系膜细胞经IL-1β,IL-6和TNFα刺激后,其培养上清中PAF的含量。结果发现,上述细胞因子均能刺激系膜细胞合成PAF,并呈一定的剂量依赖关系,IL-1β,IL-6和TNFα通过这一途径而间接地发挥病理作用,加剧肾小球损害。 n this study, the 3rd parsaged mesan-gial cells (MCs) from rat glomeruli wereseeded in a 24-well plate (5 ×105 cell/well).Then , the indicated concentration of LPS,IL-lβ, IL-6 and TNFα were added. After0. 5 h , the supernatants were collected forPAF assay. PAF was quantitated after ex-tractron and purification by thin layer chro-matography according to the aggregation ofwashed rabbit platelets as reported. The re-sult suggested that LPS, IL-lβ, IL-6 andTNFα stimulate the syhthesis of PAF byMCs in a dose-dependent manner.
