The solid-phase peptide synthesis(SPPS) of thymopentin(TP5) by Fmoc strategy

摘要 采用对碱敏感的Fmoc法α-氨基保护策略固相合成了胸腺五肽,并讨论了胸腺五肽不同固相载体、不同活化试剂对胸腺五肽固相合成的影响。试验结果表明Fmoc合成策略中各步的缩合率均在90%以上,同时对产品进行色谱分析,纯度达到83%。 Using the base-labile Fmoc group to protect α-NH 2 of amino acid, the thymopentin(TP5) has been synthesized by a solid-phase method. Then, different linkers and active reagents were investigated to optimise the synthesis of TP5 by SPPS. The experimental results showed that the ratio of the coupling cycle of each amino acid was over 90%. After cleavage of peptide resin, the purity of rude TP5 was about 83% by HPLC analysis.
