摘要
目的建立RP-HPLC法测定人血清中头孢泊肟浓度。方法用AgilentZORBAx×SBC18柱,流动相为水:乙腈:三乙胺(100:14:1,V/V/V),紫外检测波长为240nm,乙酰苯胺为内标。结果头孢泊肟保留时间在8min左右,浓度线性范围在0.25~6.12mg·L-1之间。血清中回收率平均为83.4%。结论该法准确、简便,可用作人血清中头孢泊肟浓度测定方法。
Objective To establish a reverse phase high-performance liquid chromatography (RPHPLC) method to determine the concentration of cefpodxime in human serum. Methods Separation was achieved on the Agilent ZORBA×SB-C18 column(4. 6×150 mm, 5μm) , the mobile phase consisted of water: acetonitrile :triethylamine (100:14:1,V/V/V). The UV-detection wavelength is 240 nm,acetanilide as internal standard. Results The retention time of cefpodxime was about 8 min. the calibration curves were linear in the range of 0. 25-6. 12 mg·L-1. The method recovery rate in serum was 83. 4%. Conclusion The HPLC method can be used to determine of the concentration of cefpodxime in human serum.
