应用RPSB杂交法进行微量标本中基因扩增检测

摘要 采用重复序列PCR将引物间多种基因序列目标“放大”,继而通过狭缝杂交,用特异基因探针在微量标本中可检测基因的扩增。采用本法对20份食管癌家族标本进行c-myc基因扩增检测,其结果与双PCR法所得一致。 Genomic sequences are amplified,together with a variety of inter-primer gene targets,from a smallamount of samples by repeats PCR. Amplifications of gene(s)were then detected in the amplified productswith specific gene probe (s) by slot blot hybridization. The amplification of c-myc gene was detected among20 specimens from esophageal cancer families with the present method. The result is similar to that obtainedwith double PCR.
