摘要
在实验室阶段,考察了多种无机盐和微量元素对乙酰乳酸脱羧酶发酵活力的影响,通过单因子实验确定出无机盐和微量元素的最佳产酶浓度。由于微量元素对酶活影响显著,通过回归方法计算出微量元素的理论最佳产酶浓度,并用响应面分析方法确定微量元素的最佳配比。结果为:影响较大的无机盐种类和浓度为NaCl0.25g/L,K2HPO40.003g/L,微量元素配比为Mg2+0.022g/L,Mn2+0.00076g/L,Zn2+0.00296g/L。
In small scale expriment, the effect of inorganic salts and trace element on the fermentation of α-Acetolactate decarboxylase (ALDC) was studied, and the optimum enzyme-producing condition was determined through single-factor experiment. Because of the obvious effect of trace element on ALDC activity, the optimum concentrations of trace elements were calculated through regressive method and the optimum proportion of trace elements were determined by response surface method. The result showed that: NaCl 0.25 g/L, K2HPO4 0.003 g/L, Mg2+ 0.022 g/L, Mn2+ 76 mg/L, Zn2+ 2.96 mg/L. The optimized proportion increased the enzyme activity by 3 times.
