门静脉内注射葡聚糖微球制成猪门静脉高压症模型 被引量:8

Establishment of a new pig model of portal hypertension by intraportal injection of dextran microspheres

摘要 目的复制一种肝内窦前性门脉高压模型。方法8头猪每5d注射1次葡聚糖微球,共4次。在门静脉栓塞的前后分别行门静脉造影、肝功能检查、门脉压力测定及肝脏病理检查。结果门脉造影显示葡聚糖微球栓塞门静脉末梢分支。门静脉压力栓塞前为(9.33±0.33)cmH2O,最后1次栓塞后1周为(24.17±2.24)cmH2O(1cmH2O=0.098kPa)。肝功能检查基本正常。肝脏病理示有轻度肝纤维化,同时可见异物肉芽肿形成。结论猪门静脉多次注射葡聚糖微球可以形成肝纤维化和门脉高压症模型。 Objective To create a presinusodial chronic portal venous (PV) hypertension model by means of repeated portal vein embolization.Methods Eight pigs were PV embolized with dextran microsphere once every 5 days,4 times.Portography,liver function tests,pressure measurement in the PV before and after PV embolization,and histopathologic examination of the livers were performed.Results Transhepatic portal venography performed after each embolization revealed diffuse PV branch occlusion in all cases.The mean PV pressure was increased form (9.33± 0.33) cm?H2O before embolization to (24.17± 2.24) cm?H2O one week after the last embolization (P< 0.01).Liver function tests were approximately normal.Histopathologic examination of the liver showed,in multiple PV branches,central plugs of dextran microspheres with granuloma.The liver parenchyma created slight hepatic fibrosis.Conclusion Embolizations of PV with dextran microsphere could create portal hypertension or induce hepatic fibrosis.
