摘要
以播娘蒿成熟期角果为材料,采用SMART技术构建了其全长cDNA文库。该文库的原始滴度为1.49×106pfu/ml,重组率为91.42%,扩增后文库滴度为1.15×109pfu/ml,随机挑取10个噬菌斑,PCR扩增插入片段,其长度为800~2000bp,表明所构建的文库达到了目的基因分离和克隆的要求。
By using SMART~ TM technology, a full-length cDNA library of Descurainia sophia silique was constructed. The results indicated the titer of the unamplified cDNA library was about 1.49× 10~6 pfu/ml and the recombinant rate of the library was 91.4%, the titer of the amplified library was 1.15× 10~9 pfu/ml. Ten clones from the library were chosen randomly for screening cDNA inserts by using PCR. The length of insertion fragments ranged from 800 bp to 2 000 bp, which indicated that the cDNA library was of high quality for cloning target genes.
