摘要
研究并建立了饲料中维吉尼亚霉素M1含量的高效液相色谱测定方法。样品用甲醇提取,用水稀释,经C18固相萃取柱净化,在235nm波长下用二极管矩阵检测器检测。色谱柱:AgilentZORBAXSBC18柱(4.6mm×150mm,5μm);流动相:乙腈∶水=40∶60(体积比)。维吉尼亚霉素M1质量分数在1.0~20.0μg/ml范围内与峰面积呈良好的线性关系,回收率为91.5%~94.6%,变异系数为1.7%~3.6%。
A method was developed for detecting virginiamycin M 1 in feed by HPLC with DAD detection at 235 nm. Virginiamycin M 1 was extracted from feed using methanol. The extract was diluted with water and cleaned by C 18 solid -phase extraction cartridge. For analysis, a Agilent ZORBAX SBC 18( 4.6 mm× 150 mm, 5 μm) was used with 60∶40 water-acetonitrile as mobile phase. The peak area response was linear for virginiamycin M 1 between 1.0- 20.0 μg/ml. The lowest detection limit of the method was 0.5 μg/ml, recovery was 91.5%- 94.6% and CV was 1.7%-3.6%.
