摘要
目的研究不同时期慢性阻塞性肺疾病(COPD)患者外周血中性粒细胞(PMN)凋亡、bcl2基因表达及肿瘤坏死因子α(TNF-α)、粒细胞集落刺激因子(GCSF)、白介素8(IL-8)及其相互关系。方法选择COPD患者39例,其中急性发作期20例,稳定期19例,健康成年人16例作为健康对照组,抽取静脉血5ml,肝素抗凝,分离PMN,流式细胞仪检测PMN凋亡率及bcl-2的表达率,同时用ELISA方法检测TNF-α、GCSF、IL-8的水平。结果COPD患者PMN凋亡率明显低于健康对照组,且急性发作期又明显低于缓解期,bcl-2表达与PMN凋亡呈负相关。TNF-α、GCSF、IL-8水平与PMN凋亡呈负相关。结论PMN凋亡延迟,PMN存活时间延长和持续激活与COPD的发病有关,bcl-2基因表达上调可能是COPD发病的分子机制之一,TNF-α、GCSF、IL-8可能抑制PMN凋亡。
Objective To study the changes and interrelation of polymorphonuclear neutrophils(PMN) apoptosis,the expressions of bcl-2,plasma cytokine,tumor necrosis factor(TNF),granulocyte colony-stimulating factor(G-CSF),interleukin 8(IL-8) in different period on chronic obstructive pulmonary disease(COPD) patients,and to discuss its effects during COPD occuring.Methods 39 COPD patients were chosen,20 patients on acute episode period and 19 patients on stationary phase,16 healthy adults as control group.5ml venous blood was taken and anti-coagulate with heparin,then separated the PMN,detected the rate of PMN apoptosis with flow cytometer,then detected the levels of TNF-α,G-CSF,IL-8 with ELISA methods.Results The rate of PMN apoptosis in COPD patients reduced greatly than that of control group.Furthermore,rate of PMN apoptosis in acute episode period was lower than that of stationary phase,on the contrary,the expression of blc-2 gene was negative correlation with PMN apoptosis.TNF-α,G-CSF,IL-8 was negative correlation with PMN apoptosis.Conclusion The time of PMN apoptosis delaied in COPD patients and the stock time of PMN delaied and continual activation was related with the occurrence of COPD,and the PMN apoptosis might be depressed by the TNF-α,G-CSF,IL-8.
