摘要
目的建立肠球菌体外药敏试验的流式细胞荧光法。方法以碘化丙啶(PI)作死体细胞染料,用流式细胞术检测1株肠球菌标准株和30株肠球菌临床菌株对万古霉素的敏感性。根据细菌培养物在不同浓度药物作用后所检测到的荧光强度来判断细菌的存活率,推断其MIC值,并与琼脂稀释法检测结果进行比较。结果31株肠球菌的流式细胞荧光法药敏结果与琼脂稀释法结果一致。结论流式细胞荧光法对肠球菌耐药性的体外监测具有快速、准确、客观等特点,有推广价值。
Objective To establish a method of flow cytofluorometric(FCM) antibiotic susceptibility test for Enterococcus in vitro.Methods The sensitivity to vancomycin of one standard strain and 30 isolates of Enterococcus were tested with FCM by using propidium iodide(PI) as a fluorescent probe.The survival rates of the bacteria in the culture after treatment with different dosages of vancomycin were determined according to the fluorescence strength.The MIC value of vancomycin against the 31 strains was judged with FCM,compared with agar dilution method.Result The vancomycin-susceptibility results of 31 strains of Enterococcus in FCM were correspond to that by agar dilution method.Conclusion The established method of FCM for Enterococcus is suitable for detecting bacterial drug-sensitivity,which is more rapid,accurate,and objective.
