丹参、银杏叶提取物伍用对肝纤维化大鼠病理变化的影响

Effect of Dan-shen combined with extract of Ginkgo Biloba Leaf on the pathological change of experimental hepatic fibrosis in rats

摘要 目的观察丹参、银杏叶提取物伍用对实验性肝纤维化大鼠肝脏病变的影响。方法将丹参浓缩液与银杏叶提取物用量(含生药量)分别在2-8g/kg、1.5-6g/kg范围内配伍给慢性肝纤维化大鼠灌胃。50d后,计算大鼠肝重/体重,观察其肝脏病理结构变化,部分标本观察肝细胞超微结构的变化。结果二药合用能使肝细胞变性、坏死及结缔组织增生减轻,可减轻超微结构的病理变化,且具有剂量依赖性,丹参与银杏叶提取物合用比等量丹参单用药效更明显。结论丹参、银杏叶提取物伍用可有效地延缓或逆转肝纤维化的形成,该作用呈剂量依赖性,尤以中剂量组疗效优于单用丹参组,表明二药伍用合理。 Objective To investigate the effect of Dan-shen combined with extract of Ginkgo Biloba Leaf on hepatic fibrosis of experimental rats. Methods Concentrated solution of Dan-shen ranged from 2 g/kg to 8 g/kg (equal to raw herb) combined with extract of Ginkgo Biloba L ranged from 1.5 g/kg to 6 g/kg(equal to raw herb) were administered to chronic hepatic fibrosis rats . The rats' ratios of net hepatic and body weight were calculated. The pathological change of all the samples and the ultra micro-structure of some samples were observed within 50 days. Results The hepatocyte degeneration, necrosis and the degree of connective tissue proliferation could be reduced as the two agents matched, the hepatic pathological changes of ultra micro-structure were reduced as well. The effect relied on the dosage administered, and which of Dan-shen combined with extract of Ginkgo Biloba L was more notable than the former administered singly (P< 0. 05) . Conclusion All the results confirm that Dan-shen combined with extract of Ginkgo Biloba L can delay or inverse hepatic fibrosis formation effectively, but the effectiveness depends on the dosage administered and efficacy of treatment of rats by middle dose mixture is superior to that by equal-dose Dan-shen, which can be concluded that the two agents are properly matched .
