摘要
目的了解血小板衍化生长因子(PDGF)在中枢神经损伤及修复中的作用。方法制备小白鼠颅脑损伤模型,分别在伤后1、4、7和14d处死,采用原位杂交和免疫组化染色法检测脑组织中的PDGFA、B链mRNA及其β受体的表达。结果伤后1d,PDGFA链mRNA的表达较对侧脑组织增加,持续到第4d,第7d达高峰,14d后略有下降;PDGFB链mRNA的表达于伤后第1d较对侧脑组织增加,第4d较第1d略有下降,7d后达到高峰,至14d后仍持续在高峰状态。PDGFβ受体的表达在伤后第1d较对侧脑无明显变化,第4d有所增加,第7d达高峰,14d略有下降。结论血小板衍化生长因子参与了中枢神经损伤的修复过程。
Objective To study the effects of platelet-derived growth factor (PDGF) on the injury and repair of central nervous system(CNS).Methods Mice were used to establish the local cerebral injury model and were executed on 1, 4, 7 and 14 days post-injury. The expression of PDGF-A, B chains mRNA and the β receptor of cerebral tissue were detected by in situ hybridization and immunohistochemistry, respectively.Results The expression of PDGF-A and B chains mRNA increased on 1 day after local cerebral cortex injury, and reached the highest level from 7 day to 14 day. The staining of PDGF-β receptor related inmmunoreactivity showed enhancement on 4 day post-injury, reached the highest on 7 day, dropped a little on day 14.Conclusion PDGF is involved in the repairing of CNS injury.
