纹状体边缘区学习记忆相关抑制性信号转导过程与蛋白磷酸酶1的参与作用

摘要 目的:明确蛋白磷酸酶1抑制剂岗田酸对大鼠纹状体边缘区相关的学习记忆的作用。方法:实验于2004-03/08在南方医科大学珠江医院神经生物研究室进行,选取成年SD大鼠35只,经Y迷宫训练筛选合格动物28只,随机分为正常对照组(n=6)、正常训练组(n=7)、岗田酸组(n=8)、生理盐水组(n=7)4组。正常对照组大鼠不接受任何训练,其余各组大鼠在Y迷宫中接受3d厌暗逃避反射训练,3d后岗田酸组双侧侧脑室分别缓慢注射岗田酸0.8μL(40g/L),生理盐水组注射等容积生理盐水。采用免疫组织化学染色观察了纹状体边缘区蛋白磷酸酶1、磷酸化的cAMP反应元件结合蛋白表达的改变。结果:28只大鼠进入结果分析。各组大鼠边缘区均有蛋白磷酸酶1表达,且正常对照组与正常训练组脑内边缘区蛋白磷酸酶1表达无明显差别(47.33±3.33,46.38±2.77,P=0.567);正常训练组边缘区有磷酸化的cAMP反应元件结合蛋白表达而正常对照组边缘区无表达;岗田酸组在术后12h边缘区仍有磷酸化的cAMP反应元件结合蛋白表达而生理盐水组则已经无表达。结论:纹状体边缘区学习行为相关蛋白转录调节中有蛋白磷酸酶1的参与。 AIM:To make clear the effect of protein phosphatase 1 inhibitor okadaic acid on the learning and memory in the marginal division of rat's striatum. METHODS:The experiment was carried out in the neurobiological research room of Zhujiang Hospital of Nanfang Medical University from March to August 2003.Thirt y five adult SD rats were selected,and 28 qualified rats were screened after Y maze test,and then they were randomly divided into normal control group(n=6),n ormal training group(n=7),okakaic acid group(n=8) and saline group(n=7).Rats in the normal control group received no training,and those in the other three group s were trained with 3 day escaping reflex caused by dislike of darkness in Y ma ze.After 3 days,0.8 μL okakaic acid(40/L) and saline of the same volume were in jected slowly to bilateral brain ventricles of rats in the okadaic acid injectio n group and saline group respectively.The changes of the expressions of protein phosphatase 1 and phosphorylated cAMP responsive element binding protein in the marginal division of striatum were observed with immunohistochemical staining. RESULTS:Twenty eight rats were involved in the analysis of results.Expression of protein phosphatase 1 was observed in the marginal division of striatum in a ll the groups,and there was no obvious difference between the normal control gro up and normal training group(47.33±3.33,46.38±2.77)(P=0.567).Phosphorylated cA MP responsive element binding protein expressed in the marginal division of stri atum in the normal training group but not in the normal control group;Twelve hou rs after treatment,expression of phosphorylated cAMP responsive element binding protein still existed in the okakaic acid group but no expression was observed i n the saline group. CONCLUSION:Protein phosphatase 1 is involved in the adjustment of protein tran scription related to learning behavior in the marginal division of striatum.
