摘要
目的观察海洋贝类提取物(EMS)对大鼠血浆及主动脉壁中血栓素B2(TXB2)、6-酮-前列腺素F1α(6-Keto-PGF1α)水平的影响以及EMS对脉动血栓形成时间的影响,以探讨EMS抗动脉粥样硬化(AS)的作用机制。方法将大鼠随机分组,药物均经灌胃给药。采用放射免疫法(RIA)测定各组大鼠血浆及主动脉壁中。TXB2,6-Keto-PGF1α的含量;采用电刺激损伤法测定大鼠颈动脉血栓形成时间,观察EMS的抗血栓作用。结果EMS5.4,10.8g·kg-1(相当于鲜贝)可明显降低大鼠血管壁中TXB2含量(P<0.05);10.8g·kg-1可减少血浆中TXB2水平(P<0.05);EMS对血管壁及血浆中6-Keto-PGF1α无明显影响(P>0.05);EMS10.8g·kg-1可延长大鼠颈动脉血栓形成时间(P<0.05)。结论EMS能降低血浆及血管壁中血栓素A2水平,具有调节TXA2与前列环素平衡、抑制血栓形成的作用。
OBJECTIVE: To investigate the influence of the comprehensive extract from marine shellfish (EMS) on the content of thromboxane B2(TXB 2, metabolite of thromboxane A2), 6-Keto-PGF 1α in plasma and aortic wall. To study the effect on the time of arterial thrombosis and investigate the mechanism of anti-atherosclerotic action. METHODS: The concentrations of TXB2 and 6-Keto-PGF 1α in plasma and aortic wall were determined by radio immunity analyse(RIA). The experimental carotid artery thromobosis in rat model was evaluated by electric stimulation injury. Then the activity of anti-thromobosis of EMS was investigated. RESULTS: EMS(5.4, 10.8 g·kg-1) significiantly decreased the amount of TXB2 in aortic wall (P 2 in plasma(P -1) didn't significantly effect on the concentration of 6-Keto-PGF1α (P > 0.05). EMS (10.8 g·kg-1) obviously prolonged the thromobosis time in rats(P 2 in plasma and aortic wall and the inhibition of thrombosis.
