摘要
目的研究大黄柳叶醇提取物及其黄酮类成分对喂饲高脂食料而引起的小鼠肥胖的抑制作用。方法用添加大黄柳叶醇提取物的高脂肪食料喂饲小鼠10周,测量小鼠体重、子宫周围脂肪组织重量、肝脏中三酰甘油的含量及大黄柳叶醇提取物对小肠刷状缘囊吸收脂肪酸的影响;检测从大黄柳叶中分离得到新的黄酮化合物地奥亭7-O-β-D-吡喃木糖(1→6)-β-D-吡喃葡萄糖苷在脂肪细胞中对去甲肾上腺素诱导脂肪分解的作用。结果大黄柳叶醇提取物可以抑制喂饲高脂食料引起的肥胖和脂肪肝。其主要活性成分为黄酮化合物地奥亭7-O-β-D-吡喃木糖(1→6)-β-D-吡喃葡萄糖苷,能够促进去甲肾上腺素诱导的脂肪分解,大黄柳叶醇提取物及单体化合物均无抑制胰脂肪酶活性的作用。结论大黄柳叶的减肥作用可能是由于大黄柳叶中含有促进脂肪分解作用的黄酮化合物地奥亭7-O-β-D-吡喃木糖(1→6)-β-D-吡喃葡萄糖苷及其他黄酮成分所致。
OBJECTIVE: To clarify preventing obesity effect of the alcohol extract and flavoniod constituents from leaves of Salix raddeana Laksch induced by oral administration of a high-fat diet in mice. METHODS: High fat diet-induced obese mice were treated with the alcohol extract and flavoniod constituents from leaves of Salix raddeana Laksch for 10 weeks. The weight of the body, paranietrium adipose tissue of mice and the content of triolein of the liver were measured. The effect of the new flavonol glycoside doismetin 7-O-β-D-xylopyranosyl (1&rarr6)-β-D-glucopyranoside on noradrenaline-induced lipolysis was examined with isolated fat cells. RESULTS: The extract of Salix raddeana Laksch prevented obesity and fatty liver induced by oral administration of a high-fat diet in mice. The active substance in the alcohol extract of Salix raddeana Laksch was identified as a new flavonal glycoside, which enhanced norepinephrine-induced lipolysis in fat cells. The alcohol extract and flavonoid consituents didn't inhibit the activity of pancreatic lipase. CONCLUSION: It is demonstrated that the anti-obesity effects of the leaves of Salix raddeana Laksch on high-fat diet-treated mice may be due partly to the enhancing effects of doismetin 7-O-β-D-xylopyranosyl (1&rarr6)-β-D-glucopyranoside on norepinephrine-induced lipolysis in adipose tissue.
