猪传染性胃肠炎病毒在ST细胞中增殖规律的研究 被引量:18

摘要 通过孔板培养系统以及微载体和转瓶培养系统,研究了感染复数(MOI)、病毒感染时间(TOI)和病毒维持液对细胞生长和猪传染性胃肠炎病毒(TGEV)繁殖的影响。结果表明,MOI影响细胞生长速率和单位细胞病毒产量,TOI影响细胞对病毒的敏感程度以及胞内病毒的增殖速率;丰富的维生素、氨基酸等物质有利于病毒效价的提高;采用微载体和生物反应器系统生产TGEV,病毒效价可达11.3lgTCID50/0.2mL,比方瓶培养系统提高约100倍。 Multiplicity of infection (MOI), time of infection (TOI) and medium composition that affect viral infection and replication efficiency were investigated in plate or on microcarrier in spinner bottle for development of high efficient processes of viral vaccineproduction. The effect of MOI on the cell growth rate and the virus productivity per cell was found based on understanding of parameters ruling cell-virus ~interactions . The number of viable cells at the point of infection, sensitivity of cells to virus and virus productivity per cell were confirmed to be highly dependent on the TOI. The viral titer was increased by adding vitamins or amino acids in medium. The titer of virus produced by the cells cultured on microcar-~riers in a bioreactor could reach 11.3 lgTCID_~50 /0.2mL, which was about 100 times as higherasthat produced in flasks.
