摘要
对1日龄非免疫鸡分别接种马立克病病毒(MDV)Ⅰ型国际标准强毒GA株、国内标准强毒京1株及Ⅰ型MDV疫苗毒CVI988株后第5d起,用改进的TRAP法和Bandscan软件测定分析、计算感染过程中鸡外周血淋巴细胞(PBMC)的端粒酶活性。结果,自接种MDVⅠ型强毒GA株和京1株后,在鸡尚未出现临床症状和可视病变之前端粒酶就可被检测到,并且随着感染鸡日龄的增加逐渐升高,分别在20~25日龄时其相对活力达到最高值;接种MDVCVI988后,整个试验期端粒酶活性均呈阴性。试验结果表明,MDVCVI988疫苗株具有较可靠的免疫效果,PBMC端粒酶活性与马立克病的发病进程具有一定的相关性。
One-day-old unvaccinated chickens were inoculated with standard MDVⅠ virulent strain GA, Jing-1 and attenuated nononcogenic vaccine strain CVI988. From day 5 post-inoculation(PI) the ~telomerase activities of peripheral blood mononuclear cells(PBMC) were detected during the inoculated ~period by using the modified telomerase repeat amplification protocol assay and software of Bandscan. The ~results showed that the telomerase in lymphocyte began to be detected before appearance of clinical symptoms and visible pathological changes among the samples infected with virulent strain GA or Jing-1 and the telomerase activities were gradually getting high as the days PI. Telomerase activities got their peak on day 20 and 25 PI respectively. No telomerase activities in chickens inoculated with serotype 1 vaccine CVI988 were measured throughout the experimental period. The study indicated that the attenuated nononcogenic vaccine strain MDVⅠ CVI988 had reliable immune results and the telomeraseactivities in lymphocyte are correlated with the stages of MD tumor.
