甘蓝型油菜MICMS恢复基因的RAPD标记 被引量:6

摘要 以MICMS育性分离群体(宁A6/宁R1)F2为基础群体,结合BSA法,利用500条10bp的随机引物对不育与可育DNA池进行筛选,存在差异的引物再对分离群体进行检测,获得了与MICMS恢复基因Rfm连锁的RAPD标记2个,即:OPH1590和OPS7380。他们位于恢复基因的两侧,遗传图距分别为5.6cM和17.3cM。 The fertility segregating population (Ning A6/Ning R1)F_2 consisted of 159 individual plants was used in a bulked segregation analysis (BSA) for the identification of molecular markers linked to fertility restoration gene of MI CMS. Five hundred random primers were screened for polymorphic bands between the two DNA pools of male fertile and the male sterile plants, and primers generated polymorphic bands were test in the whole population. Two RAPD markers, OPH1_(590) and OPS7_(380), were identified to be linked to the restorer gene Rfm and located to each side of the restorer gene with genetic distances of 5.6cM and 17.3cM, respectively.
